Background

USP14 (Ubiquitin Specific Peptidase 14) is a deubiquitinating enzyme that serves as a critical regulatory subunit of the 26S proteasome, functioning primarily at the interface between the 19S regulatory particle and the 20S core particle to modulate protein degradation. By reversibly removing ubiquitin chains from substrate proteins prior to their entry into the proteolytic core, USP14 acts as a "gatekeeper" that can either rescue substrates from degradation or fine-tune the rate of proteolysis, thereby playing a pivotal role in maintaining cellular protein homeostasis and regulating key physiological processes such as DNA damage response, oxidative stress management, and immune signaling. Due to its ability to stabilize oncoproteins and influence tumor cell survival, USP14 has emerged as a significant therapeutic target in cancer research, where its inhibition is being explored to enhance the degradation of disease-causing proteins and sensitize tumors to chemotherapy, highlighting its dual potential as both a protector of cellular integrity and a driver of pathological states when dysregulated.
