Background
Urokinase-type plasminogen activator (uPA) is a 411-residue, trypsin-like serine protease that converts the abundant zymogen plasminogen into the broad-spectrum protease plasmin; by binding to its high-affinity glycolipid-anchored receptor uPAR on the plasma membrane, uPA focuses pericellular proteolysis to the cell surface, thereby driving physiological extracellular-matrix remodeling during wound healing and tissue involution as well as pathological processes such as tumor cell invasion, angiogenesis and metastasis, while its non-proteolytic, growth-factor-like amino-terminal fragment (ATF) triggers intracellular signaling cascades that promote cell migration, proliferation and survival, making the uPA/uPAR system a key therapeutic target in cancer, fibrosis and thrombosis.
