Background
UQCRC2 (ubiquinol-cytochrome c reductase core protein 2), encoded on chromosome 16p12.2, is a 453-amino-acid, 48.4 kDa integral component of mitochondrial complex III (cytochrome b-c1 complex) that stabilizes the ~450 kDa multisubunit transmembrane assembly and, by facilitating electron flow from ubiquinol to cytochrome c coupled to proton pumping across the inner membrane, powers the electrochemical gradient essential for oxidative phosphorylation, while its peptidase-M16-family processing activity contributes to maturation of the Rieske iron-sulfur protein and mutations in UQCRC2 are linked to mitochondrial complex III deficiency nuclear type 5.

IHC shows positive staining in paraffin-embedded human liver. Anti-UQCRC2 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HepG2 cells. Anti-UQCRC2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
