Background

The kit employs homogeneous time-resolved fluorescence (TR-FRET) technology to measure the interaction between the DDB1/CRBN complex and VAV1 mediated by the molecular glue MRT-6160 or test compounds. This method provides a high-throughput format for the simple and rapid detection of small molecules capable of mediating the interaction between the DDB1/CRBN complex and VAV1.

As shown, the interaction between DDB1/CRBN and VAV1 is detected using a Eu-labeled anti-Tag1 antibody (TR-FRET donor) and an Accepter-labeled anti-Tag2 antibody (TR-FRET acceptor). Due to the molecular glue MRT-6160 mediating the interaction between DDB1/CRBN and VAV1, the donor and acceptor antibodies are brought into close proximity. Excitation of the donor antibody triggers fluorescence resonance energy transfer (FRET) to the acceptor antibody, causing the acceptor antibody to specifically emit a signal at a wavelength of 665 nm. This specific signal is proportional to the extent of the interaction mediated by MRT-6160 between DDB1/CRBN and VAV1. This homogeneous assay is simple to perform and requires no washing steps.
