Background
This kit utilizes Homogeneous Time-Resolved Fluorescence(TR-FRET) technology to measure the interaction between IL21 and IL21R. Thismethod enables simple and rapid high-throughput screening of inhibitors andantibody blockers.
As illustrated, the interaction between IL21 and IL21R isdetected using an Eu-labeled anti-Tag1 antibody (TR-FRET donor) and anAcceptor-labeled anti-Tag2 antibody (TR-FRET acceptor). The binding of IL21 toIL21R brings the donor and acceptor antibodies into proximity. Excitation ofthe donor antibody then triggers Fluorescence Resonance Energy Transfer (FRET)to the acceptor antibody, resulting in a specific emission signal at 665 nm.The intensity of this specific signal is proportional to the extent of theIL21/IL21R interaction. This homogeneous assay is simple to perform andrequires no washing steps.
