Background
This kit uses Homogeneous Time-Resolved Fluorescence(TR-FRET) technology to measure the interaction between IL17A and IL17RA. Thismethod enables simple and rapid high-throughput screening of inhibitors andantibody blockers.
As shown in the figure, the interaction between IL17A andIL17RA is detected using an Eu-labeled anti-Tag1 antibody (TR-FRET donor) andan Acceptor-labeled IL17A protein (TR-FRET acceptor). The binding of IL17A toIL17RA brings the donor antibody and the acceptor-labeled protein into closeproximity. Excitation of the donor antibody then triggers FluorescenceResonance Energy Transfer (FRET) to the acceptor, resulting in the emission ofa specific signal at 665 nm. The intensity of this specific signal isproportional to the extent of the IL17A/IL17RA interaction. This homogeneousassay is simple to perform and requires no washing steps.
