Background
This kit utilizes HomogeneousTime-Resolved Fluorescence (TR-FRET) technology to measure the interactionbetween IL10A and IL10RA. This method enables simple and rapid high-throughputscreening of inhibitors and antibody blockers.
As illustrated, the interactionbetween IL10A and IL10RA is detected using an Eu-labeled Anti-Tag1 antibody(TR-FRET donor) and an Acceptor-labeled IL10A protein (TR-FRET acceptor). Thebinding of IL10A to IL10RA brings the donor antibody and the acceptor proteininto close proximity. Excitation of the donor antibody then triggersFluorescence Resonance Energy Transfer (FRET) to the acceptor antibody,resulting in the emission of a specific signal at 665nm. The intensity of thisspecific signal is proportional to the extent of the IL10A and IL10RAinteraction. This homogeneous assay is simple to perform and requires nowashing steps.
