Background
This kit utilizes Homogeneous Time-Resolved Fluorescence(TR-FRET) technology to measure the interaction between BRD4 and the[LYS(5,8,12,16)AC] H4(1-21) peptide. This method enables simple and rapidhigh-throughput screening of inhibitors and blocking antibodies.
As illustrated, the interaction between BRD4 and the[LYS(5,8,12,16)AC] H4(1-21) peptide is detected using a Eu-labeled Anti-Tag1antibody (TR-FRET donor) and an Acceptor-labeled Anti-Tag2 antibody (TR-FRETacceptor). The binding of BRD4 to the [LYS(5,8,12,16)AC] H4(1-21) peptidebrings the donor and acceptor antibodies into close proximity. Excitation ofthe donor antibody then triggers Fluorescence Resonance Energy Transfer (FRET)to the acceptor antibody, resulting in a specific emission signal at 665 nm.The intensity of this specific signal is proportional to the degree ofinteraction between BRD4 and the [LYS(5,8,12,16)AC] H4(1-21) peptide. Thishomogeneous assay is simple to perform and requires no washing steps.
