Background
This kit utilizes Time-ResolvedFluorescence Resonance Energy Transfer (TR-FRET) technology to measure theinteraction between GSPT1 and CRBN mediated by the molecular glue CC-885 andtest compounds. This method provides a simple, rapid, and high-throughputformat for detecting small molecules capable of mediating GSPT1-CRBNinteractions.
As shown, the interaction betweenGSPT1 and CRBN is detected using an anti-Tag1 antibody labeled with Eu (TR-FRETdonor) and an anti-Tag2 antibody labeled with Ac (TR-FRET acceptor). Since themolecular glue CC-885 mediates the interaction between GSPT1 and CRBN, thedonor and acceptor antibodies are brought into proximity. Excitation of thedonor antibody triggers fluorescence resonance energy transfer (FRET) to theacceptor antibody, resulting in specific emission of a 665 nm signal. Thisspecific signal is proportional to the degree of interaction between CC-885,GSPT1, and CRBN. This homogeneous assay is simple to perform and requires nowash steps.
