Store the kit at -80°C or below.When stored properly, the reagents remain stable until the expiration dateprinted on the label. Once thawed, the CD40 and CD40L stock solutions canundergo one additional freeze-thaw cycle; the labeled antibody solutions canundergo one additional freeze-thaw cycle. To avoid repeated freeze-thaw cycles,it is recommended to aliquot the remaining stock solutions into single-usecontainers for storage at -80°C. Thawed dilutions and Detection Buffer can bestored at 2-8°C. |
Background
This kit utilizes HomogeneousTime-Resolved Fluorescence Resonance Energy Transfer (TR-FRET) technology tomeasure the interaction between CD40 and CD40L. This method enables simple andrapid high-throughput screening of inhibitors and blocking antibodies.
As illustrated, the interactionbetween CD40 and CD40L is detected using an Eu-labeled Anti-Tag1 antibody(TR-FRET donor) and an Acceptor-labeled Anti-Tag2 antibody (TR-FRET acceptor).The binding of CD40 to CD40L brings the donor and acceptor antibodies intoproximity. Excitation of the donor antibody then triggers FRET to the acceptorantibody, resulting in a specific emission signal at 665 nm. The intensity ofthis specific signal is proportional to the degree of CD40/CD40L interaction.This homogeneous assay is simple to perform and requires no washing steps.
