Store the kit at -80°C or below. When storedappropriately, the reagents remain stable until the expiration date marked onthe label. Once thawed, BCL6 and CRBN stock solutions can undergo oneadditional freeze-thaw cycle; labeled antibody solutions can undergo oneadditional freeze-thaw cycle. To avoid repeated freeze-thaw cycles, it isrecommended to aliquot the remaining stock solutions into single-use containersfor storage at -80°C. Thawed diluents and Detection Buffer can be stored at2-8°C. |
Background
This kit uses Homogeneous Time-Resolved Fluorescence(TR-FRET) technology to measure the molecular glue ARV-393 and testcompound-mediated interaction between the DDB1/CRBN complex and BCL6. Thismethod enables simple and rapid high-throughput screening of small moleculescapable of mediating the interaction between the DDB1/CRBN complex and BCL6.
As shown, the interaction between DDB1/CRBN and BCL6 isdetected using a Eu-labeled Anti-Tag1 Antibody (TR-FRET donor) and anAcceptor-labeled Anti-Tag2 Antibody (TR-FRET acceptor). As the molecular glueARV-393 mediates the interaction between DDB1/CRBN and BCL6, the donor andacceptor antibodies are brought into proximity. Excitation of the donorantibody triggers Fluorescence Resonance Energy Transfer (FRET) to the acceptorantibody, resulting in a specific emission signal at 665 nm. This specificsignal is proportional to the degree of interaction mediated by ARV-393 betweenDDB1/CRBN and BCL6. This homogeneous assay is simple to perform and requires nowashing steps.
