Background
This kit uses HomogeneousTime-Resolved Fluorescence (TR-FRET) technology to measure the molecular glueBCL-XL DEGRADER-2 and test compound-mediated interaction between VHL andBCL-XL. This method enables a simple and rapid high-throughput format for screeningsmall molecules that can mediate the interaction between VHL and BCL-XL.
As shown, the interaction betweenVHL and BCL-XL is detected using an Eu-labeled anti-Tag1 antibody (TR-FRETdonor) and an Acceptor-labeled anti-Tag2 antibody (TR-FRET acceptor). Since themolecular glue BCL-XL DEGRADER-2 mediates the interaction between VHL andBCL-XL, the donor and acceptor antibodies are brought into proximity.Excitation of the donor antibody triggers Fluorescence Resonance EnergyTransfer (FRET) to the acceptor antibody, resulting in a specific emissionsignal at 665 nm from the acceptor. This specific signal is proportional to thedegree of interaction mediated by BCL-XL DEGRADER-2 between VHL and BCL-XL.This homogeneous assay is simple to perform and requires no washing steps.
