Background

Tyrosinase is the key rate-limiting enzyme in the melanin synthesis pathway and the most important regulatory node in the pigmentation control network. It is a copper-containing type I transmembrane glycoprotein, with its intra-catalytic domain containing a dinuclear copper center that initiates the biosynthetic cascade from tyrosine to eumelanin or pheomelanin by hydroxylating monophenols to o-diphenols (monophenolase activity) and oxidizing o-diphenols to o-quinones (diphenolase activity). At the regulatory level, its expression is primarily controlled at the transcriptional level by MITF (microphthalmia-associated transcription factor) , while its enzymatic activity is dynamically modulated by multiple factors including pH, copper availability, and phosphorylation modifications. Precisely due to its central role in melanogenesis, Tyrosinase has become a core target for pigmentary disorders (such as albinism and vitiligo) and skin-lightening intervention strategies; meanwhile, its aberrant expression due to gene mutations is directly linked to oculocutaneous albinism type I, and its upregulation as a tumor-associated antigen in melanoma cells has also made it a popular target for melanoma immunotherapy and targeted therapy.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human skin. Anti-Tyrosinase antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
