Background
Tyrosine kinase 2 (TYK2) is a 1,187-residue Janus kinase that, by partnering with JAK1 to phosphorylate STAT1/2/3/4/5, transduces cytokine signals from IL-12, IL-23, type I IFNs and related receptors, thereby steering Th1/Th17 differentiation, antiviral and antitumor immunity; its unique pseudokinase–kinase domain architecture allows allosteric auto-inhibition, and human missense variants that weaken this brake—such as I684S, P1104A or the catalytic V362F—confer either protection against autoimmune diseases (by dampening IL-23-driven inflammation) or, conversely, risk of systemic lupus, psoriasis, multiple sclerosis and cancers, making selective TYK2 inhibitors like deucravacitinib promising therapies.
