Background

TXNRD1, or cytosolic thioredoxin reductase 1, is a homodimeric flavoprotein of approximately 649 amino acids and 71 kDa, encoded by the TXNRD1 gene and belonging to the pyridine nucleotide-disulfide oxidoreductase family. It is a core component of the mammalian thioredoxin system. Each subunit of this protein contains an FAD cofactor and a rare amino acid—selenocysteine—located at the C-terminal active center, which is encoded by the UGA codon and requires precise translation through a SECIS element. Functionally, TXNRD1 utilizes NADPH as an electron donor to primarily reduce oxidized thioredoxin, while also directly reducing various substrates such as hydrogen peroxide, thereby playing a critical role in maintaining cellular redox homeostasis and regulating cell growth and differentiation. The protein is primarily localized in the cytosol, but alternative splicing generates different isoforms targeted to the nucleus or plasma membrane, further expanding its functional diversity. In addition to its classic antioxidant functions, recent studies have revealed non-enzymatic activities of TXNRD1, such as enhancing cGAS enzyme activity by binding to it in senescent cells, thereby driving the cGAS-STING inflammatory pathway to promote the senescence-associated secretory phenotype, thus playing an important role in the tumor microenvironment and inflammaging. Given its critical role in supporting proliferation and counteracting oxidative stress in cancer cells, TXNRD1 has emerged as a promising target for anticancer drug development.
