Background
TSLP(R127A, R130S) is a full-length recombinant thymic stromal lymphopoietin stabilized by R127A and R130S double mutations, which prevent proteolytic truncation during expression by disrupting the furin cleavage site in the CD loop region (125-131 RRKRK basic cassette), thereby preserving the open four-helix bundle core (αA-αD) and dual-receptor binding activity for TSLPR/IL-7Rα; this mutant serves as a homogeneous, highly active standard antigen widely used for screening, affinity determination, and epitope mapping of anti-TSLP antibodies (e.g., Tezepelumab), as well as cross-reactivity assessment in non-human primate species such as cynomolgus monkeys, providing an indispensable molecular tool for the development of TSLP-targeted therapeutics against asthma and atopic dermatitis.
