Background

TSLP (thymic stromal lymphopoietin) is an IL-7-like cytokine produced by epithelial cells that activates the JAK/STAT signaling pathway through a ternary complex composed of its receptor TSLPR and IL-7Rα, driving dendritic cell maturation and Th2-type immune responses, and serving as a core upstream regulator of type 2 inflammation (asthma, atopic dermatitis). Due to >95% sequence homology of TSLP between cynomolgus monkeys and humans, cynomolgus monkeys have become a critical species for nonclinical safety evaluation of anti-TSLP therapeutics such as tezepelumab; their asthma models have effectively validated the in vivo efficacy of TSLP/TSLPR blockade strategies, providing irreplaceable translational medicine evidence for the development of TSLP-targeting antibody drugs.
