| Usage | 1. After slicing or chip samples until the step of incubating polyHRP secondary antibody is completed, wash PBS three times for 5min each time; 2. The concentrated fluorescent dye and the TSA buffer are uniformly mixed according to the ratio of 1:50-1: 400 to prepare the TSA fluorescent dye reaction solution; 3. Add the prepared TSA fluorescent dye reaction solution dropwise to the sections to evenly cover the tissue at room temperature for 1-15min (the best time is 5min-10min), and wash it three times with PBS (in the pre-experiment, you can first dye for 1min to wash the fluorescent dye and then observe the staining effect under a microscope. If the positive is weak, continue to add fluorescent dye dropwise to strengthen the staining intensity until the appropriate intensity, and then continue to the next step). |

TSA fluorescent dye reaction solution = TSA xxx fluorescent dye + TSA buffer; Wherein TSA xxx fluorescent dye: TSA buffer = 1: 100; The dilution ratio of TSA xxx fluorescent dye and TSA buffer can be flexibly adjusted and optimized according to specific conditions, and the optimal range is 1:50-1: 400 (1: 100 can get the best results in most cases); It is generally recommended that if the primary antibody incubation time is within 1h-3h at room temperature, the recommended dilution ratio is 1:50-1: 200, and if the primary antibody incubation time is 4 overnight, the recommended dilution ratio is 1: 200-1: 400 or higher. |
