Background

TriMethyl-Histone H3 (K36) refers to the trimethylation modification at the 36th lysine residue of histone H3, a highly conserved epigenetic mark primarily catalyzed by the SETD2 methyltransferase. It is widely distributed throughout the transcribed regions of actively transcribed genes (from downstream of the transcription start site to the gene end) and plays critical regulatory roles in transcriptional elongation, RNA splicing, and DNA damage repair. During transcription, phosphorylated RNA polymerase II recruits SETD2, which progressively catalyzes the deposition of H3K36me3 on nucleosomes. This modification subsequently regulates transcriptional elongation rates, suppresses cryptic promoter initiation, and promotes exon splicing through the recognition of effector proteins such as PSIP1/LEDGF and ZMYND11. In the field of DNA damage repair, H3K36me3 mediates efficient homologous recombination repair and mismatch repair by recruiting factors including the MSH6 mismatch repair complex and KDM4A/B. Reduced H3K36me3 levels or aberrant distribution are closely associated with various diseases, such as SETD2 mutation-driven cancers (including renal cancer and leukemia), as well as developmental abnormality syndromes caused by dysfunction of KMT3C/KMT3B (e.g., Lupski syndrome and Sotos-like phenotypes).

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
