Background

Transthyretin (TTR) is a tetramer composed of four identical subunits, with each monomer rich in β-sheets and forming an internal channel within its structure for ligand binding. It primarily functions as a transport carrier in the blood and cerebrospinal fluid, responsible for transporting thyroid hormone thyroxine (T4) and retinol (vitamin A). Genetic mutations or age-related misfolding of TTR can lead to its aggregation into amyloid fibrils, triggering a series of diseases known as transthyretin amyloidosis (ATTR), including both hereditary and wild-type amyloidosis, which can affect multiple organs such as nerves, heart, and eyes. Consequently, this protein is a key target for disease diagnosis, disease monitoring, and emerging therapies (such as stabilizing the tetrameric structure or clearing amyloid deposits).
