Background

The Transferrin Receptor (TfR), primarily existing as the homodimeric glycoprotein TfR1 (CD71), is a critical cell surface molecule responsible for regulating cellular iron uptake by binding iron-loaded transferrin with high affinity and internalizing the complex via clathrin-mediated endocytosis, a process essential for delivering iron to the cytoplasm where it supports vital functions such as DNA synthesis, oxygen transport, and mitochondrial respiration; upon iron release in the acidic endosomal environment, the apo-transferrin remains bound to the receptor and is recycled back to the cell surface for dissociation at neutral pH, while the receptor's expression is tightly post-transcriptionally controlled by the Iron Regulatory Protein (IRP) system in response to intracellular iron levels to prevent toxicity or deficiency, making it not only a fundamental component of systemic iron homeostasis but also a significant biomarker for rapidly proliferating cells like erythroid precursors and many cancers, as well as a strategic target for drug delivery systems designed to cross the blood-brain barrier.

Immunohistochemistry

Immunocytochemistry

ICC shows positive staining in K562 cells. Anti-Transferrin receptor antibody was used at 1/50 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
