Background

The TORC1 protein complex, full name Target of Rapamycin Complex 1, is a highly conserved serine/threonine protein kinase complex in all eukaryotes and serves as a central hub in the regulation of cell growth and metabolism. This complex features the protein kinase TOR (known as mTOR in mammals) as its catalytic core subunit, along with key structural subunits such as Kog1 (Raptor in mammals) and Lst8. Structurally, TORC1 typically forms a dimer with a hollow architecture, in which the kinase domain and regulatory subunits collectively constitute a sophisticated molecular machine capable of sensing and integrating diverse signals—including amino acids, energy levels, growth factors, and stress—to coordinate cellular anabolic and catabolic processes. Upon activation by upstream signals, TORC1 phosphorylates downstream effector proteins, such as ribosomal protein S6 kinase (S6K) and the translation inhibitor 4EBP1, thereby promoting anabolic processes like protein synthesis while inhibiting catabolic processes such as autophagy. Consequently, dysregulation of TORC1 function is closely associated with various pathological conditions, including cancer, metabolic disorders, and aging, making it a critical target for drug development.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded mouse cerebral cortex. Anti-TORC1/CRTC1 antibody was used at 1/4000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in Neuro-2a cells. Anti-TORC1/CRTC1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
