Background
TNF‑β, also termed lymphotoxin‑α (LTA), is a pleiotropic cytokine secreted mainly by activated T and B lymphocytes, participating in immune regulation, lymphoid organ development and inflammatory signal transduction. Its concentration can be detected by immunoassays, and serum or tissue‑level TNF‑β carries distinct clinical and research value.
Upon immune activation, TNF‑β binds to TNF receptors, inducing endothelial activation, leukocyte recruitment and pro‑inflammatory mediator release. Increased TNF‑β levels are associated with multiple autoimmune diseases, such as rheumatoid arthritis, systemic lupus erythematosus and multiple sclerosis, and its concentration partly reflects ongoing immune activation, helping assess disease activity. It also plays roles in chronic infection, allograft rejection and tumour microenvironment modulation; elevated TNF‑β may indicate aggravated immune attack in transplant recipients, while in certain cancers it shapes anti‑tumour or tumour‑promoting immune responses for prognostic reference.
However, TNF‑β lacks high diagnostic specificity. It rises in diverse inflammatory states triggered by infection, trauma and other cytokines, so it cannot serve as a standalone diagnostic marker. Standardized cut‑off values are not universally established, and large prospective cohorts are required to validate its clinical thresholds. Collectively, TNF‑β measurement offers supplementary information for evaluating lymphocyte‑mediated immune activation, stratifying inflammatory risk, monitoring immune‑related disease progression and exploring novel immunomodulatory therapeutic targets.
