Background
TLR2, also known as CD282, is an approximately 89-kDa type I transmembrane receptor of the Toll-like receptor family that plays a pivotal role in innate immunity by recognizing pathogen-associated molecular patterns (PAMPs) such as bacterial lipoproteins and cell wall components; it cooperates with co-receptors like LY96 and forms heterodimers with TLR1 or TLR6 to activate the MYD88- and TRAF6-dependent signaling pathway, leading to NF-κB activation, cytokine secretion, and inflammatory responses, while being abundantly expressed in peripheral blood leukocytes, especially monocytes, and involved in host defense against Gram-positive bacteria and yeast.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human tonsil. Anti-TLR2 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HepG2 cells. Anti-TLR2 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
