Background

TIMP1 (tissue inhibitor of matrix metalloproteinases 1) is a glycoprotein secreted by multiple cell types and a core member of the TIMP family. Its primary function is to specifically inhibit the proteolytic activity of matrix metalloproteinases (MMPs). Through chelation of the zinc ion at the active site of MMPs via its N-terminal domain, TIMP1 reversibly binds and inhibits various MMPs (such as MMP-1, MMP-3, and MMP-9) in a 1:1 stoichiometric ratio, thereby regulating the degradation and remodeling balance of the extracellular matrix. Additionally, TIMP1 possesses non-classical functions independent of MMP inhibition, including promoting cell proliferation, inhibiting apoptosis (via activation of the PI3K/AKT signaling pathway), and regulating inflammatory responses and angiogenesis. In pathophysiological processes, aberrant expression levels of TIMP1 are closely associated with various diseases: in liver fibrosis and pulmonary fibrosis, sustained high expression of TIMP1 leads to excessive matrix accumulation by inhibiting MMP activity, promoting disease progression. In the tumor microenvironment, TIMP1 is associated with poor prognosis and enhanced metastatic potential in various cancers (such as colorectal cancer, breast cancer, and liver cancer) by promoting epithelial-mesenchymal transition and inhibiting tumor cell apoptosis. Notably, TIMP1 also serves as a potential biomarker for diagnosing and monitoring the progression of fibrotic diseases and certain cancers. In summary, TIMP1, as a key regulator of extracellular matrix homeostasis, plays dual and complex functions in tissue remodeling, inflammatory regulation, and tumor progression.
