Background

Thymidine Kinase 1 (TK1) is a key enzyme in the pyrimidine nucleotide salvage pathway, responsible for catalyzing the conversion of thymidine to thymidine monophosphate (dTMP), which is essential for DNA synthesis and cell proliferation. Unlike TK2, which is primarily localized in mitochondria, TK1 is a cytosolic enzyme that exhibits cell cycle-dependent expression, with its activity sharply rising at the G1/S phase transition and peaking during S phase, followed by rapid degradation after mitosis, making it a widely used marker for cell proliferation activity. In clinical applications, serum TK1 levels have become an important biomarker for prognosis, treatment response monitoring, and recurrence assessment in various tumors (including lymphoma, leukemia, breast cancer, and lung cancer), where elevated levels typically indicate active tumor cell proliferation and disease progression. Additionally, TK1 serves as an important target for the development of antiviral and anticancer drugs; for example, nucleoside analog drugs (such as acyclovir and ganciclovir) rely on viral or cellular TK for phosphorylation-mediated activation, while highly selective TK1 inhibitors are being investigated for the treatment of hematological malignancies and autoimmune diseases. Notably, TK1 levels can also be elevated under certain normal physiological conditions such as tissue regeneration and inflammation, so clinical interpretation should be combined with other indicators for comprehensive assessment.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

ICC shows positive staining in HCT116 cells. Anti- Thymidine kinase 1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
