Background
TEL1 protein is the budding-yeast ortholog of mammalian ATM, a PIKK-family kinase recruited to DNA double-strand breaks by the Mre11-Rad50-Xrs2 (MRX) complex, where it phosphorylates multiple SQ/TQ motifs to launch the DNA-damage checkpoint, controls telomere length, stabilizes MRX at DSB ends independently of its catalytic activity to promote end-tethering and repair by both homologous recombination and non-homologous end-joining, and can be further activated through direct interaction with phosphoinositides such as PI(4)P.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon. Anti-TEL1 antibody was used at 1/100 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
