Background
TDG (thymine DNA glycosylase) is a 410-residue nuclear enzyme that belongs to the uracil DNA glycosylase (UDG) superfamily and acts as a key guardian of epigenetic stability by excising thymine, 5-formylcytosine, 5-carboxylcytosine and other deaminated or oxidized bases from G•T, G•U or G•5fC/5caC mispairs in double-stranded DNA, thereby initiating base-excision repair (BER), reshaping CpG methylation landscapes and preventing C→T transition mutations and transcriptional misregulation in embryonic stem cells and somatic tissues.

IHC shows positive staining in paraffin-embedded human testis. Anti-TDG antibody was used at 1/50 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
