Background

TBX18 is a key developmental regulatory factor belonging to the T-box transcription factor family, playing an indispensable core role in embryonic development through its highly conserved T-box DNA-binding domain. As a transcriptional repressor, TBX18 can bind to specific DNA sequences and precisely regulate the expression of downstream target genes, which is essential for the morphogenesis of multiple tissues and organs. Its function is profoundly embodied in heart development, where it is not only a cell source marker required for the formation of the epicardium and coronary vessels, but also critical for the normal development of the head region of the sinoatrial node (the heart's pacemaker). In addition, TBX18 is also involved in the development of the ureter and spine.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human tonsil. Anti-TBX18 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

IHC shows positive staining in paraffin-embedded human cervical cancer.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- TBX18 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
