Background

SYT1 is a key calcium ion sensor primarily located on synaptic vesicle membranes, playing a central role in the rapid synchronous release of neurotransmitters. Its structure contains two key C2 domains (C2A and C2B), among which the C2B domain interacts with phospholipids and the SNARE complex on the plasma membrane through its basic residues, driving the anchoring and fusion of synaptic vesicles with the presynaptic membrane. When an action potential arrives, SYT1 senses the calcium influx and triggers rapid exocytosis of vesicles, thereby ensuring the precise transmission of neural signals. Dysfunction of SYT1 is closely associated with various diseases: its pathogenic variants can cause Baker-Gordon syndrome, a neurodevelopmental disorder characterized by motor developmental delay, intellectual disability, hypotonia, and visual problems; in the field of oncology, the role of SYT1 is tissue-specific—for example, it is upregulated in hepatocellular carcinoma and promotes tumor progression, whereas in prostate cancer and colorectal cancer it is downregulated and plays a role in suppressing tumor metastasis; furthermore, SYT1 deficiency can also aggravate pressure overload-induced cardiac hypertrophy and dysfunction.
