Background

SUZ12, also known as Zeste 12 Homolog, is a protein encoded by the human SUZ12 gene and is a component of the Polycomb Repressive Complex 2 (PRC2). SUZ12 plays a significant role in epigenetic modification, particularly in Regulating tyrosine trimethylation at position 27 of histone H3. SUZ12 has been implicated in the proliferation and metastasis of various types of cancer. It is overexpressed in tumors such as lung, stomach, colorectal, and breast cancer, where it promotes tumor growth and invasion. In hepatocellular carcinoma, SUZ12 downregulation has been associated with the upregulation of tumor stem cell marker genes, suggesting its importance in tumorigenesis and cancer progression.

Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized NIH/3T3 (Mouse embryonic fibroblast) labelling SUZ12 antibody at 1/50 dilution (1 μg) / (Red) compared with a Rabbit monoclonal IgG (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti - Rabbit IgG Alexa Fluor® 488 was used as the secondary antibody.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- SUZ12 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
