Background

Survivin is a 16.5 kDa member of the inhibitor of apoptosis (IAP) family that uniquely integrates control of cell division and suppression of apoptosis by binding to caspases-3 and -7, thereby blocking the terminal execution phase of programmed cell death, while also associating with the chromosomal passenger complex at centromeres during mitosis to ensure accurate chromosome segregation; it is transcriptionally repressed by wild-type p53, strongly up-regulated in virtually every human cancer through PI3K/AKT, STAT3, β-catenin/TCF and HIF-1α pathways, and correlates with poor prognosis, resistance to chemo- and radiotherapy, and increased tumor recurrence, making it a high-value therapeutic target addressed by antisense oligonucleotides, dominant-negative mutants, small-molecule antagonists and immunotherapies that collectively seek to restore apoptotic sensitivity and inhibit unchecked proliferation in malignant cells.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-Survivin antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HeLa cells (top panel) and low staining in MCF7 cells (below panel). Anti-Survivin antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
