Background
Mouse interleukin-4 (IL-4) is an 18–20 kDa monomeric glycoprotein cytokine primarily produced by Th2 cells, mast cells, basophils, eosinophils, NKT cells, and γδT cells. As a central regulator of type 2 immunity, it drives Th2 differentiation via the JAK-STAT6 pathway upon binding to IL-4 receptor complexes (IL-4Rα paired with γc or IL-13Rα1), inducing transcription factors like GATA3 and c-Maf. Functionally, IL-4 promotes B cell proliferation, class switching to IgG1/IgE, and activation of eosinophils/mast cells, while suppressing Th1 responses. Its roles are context-dependent: IL-4 exerts protective effects in acute lung injury (reducing TNF-α/IL-6 and apoptosis) and cerebral ischemia (attenuating neuroinflammation), yet can exacerbate infections (e.g., mousepox) by suppressing cytolytic lymphocyte responses.

Stimulate 2×10⁵ cells/well of mouse spleen cells with Con A for 20 hours at 37 °C in a 5% CO₂ incubator. Repeat across 8 wells in 3 batches to evaluate inter-batch and intra-batch precision. Data presented include mean spot count, range, and coefficient of variation.

Stimulate 2×10⁵ cells/well of mouse spleen cells with Con A. After 20 hours of stimulation at 37 °C in a 5% CO₂ incubator, incubate with HRP-labeled detection antibody at room temperature for 2 hours. Add substrate for color development, followed by image analysis and spot counting.
