Background
Interleukin (IL)-5 is a cytokine dimer derived from both hematopoietic and non-hematopoietic cell lines that primarily contribute to the proliferation, differentiation, and survival of eosinophils. While other cytokines have been implicated in the maturation, survival, and function of eosinophils, as one of its primary roles includes stimulating production of these cells from the bone marrow. Given eosinophilic prominence in a variety of disease states, considerable research has focused on the development of therapeutic drugs targeting IL-5-dependent signaling.

Stimulate 2×10⁵ cells/well of cryopreservation cynomolgus macaque PBMCs with PMA and Ionomycin for 22 hours at 37°C in a 5% CO₂ incubator. Repeat across 6 wells in 3 batches to evaluate inter-batch and intra-batch precision. Data presented include mean spot count, range, and coefficient of variation.

Stimulate 2×10⁵ cells/well of cryopreserved cynomolgus macaque PBMCs with PMA and Ionomycin. After 22 hours of stimulation at 37°C in a 5% CO₂ incubator, incubate with HRP-labeled detection antibody at room temperature for 2 hours. Add substrate for color development, followed by image analysis and spot counting.
