Background
STAT6 (Signal Transducer and Activator of Transcription 6) is a transcription factor that plays a pivotal role in the cellular response to interleukin-4 (IL-4). It is activated through IL-4-induced tyrosine phosphorylation. Its primary function is to mediate most of IL-4's effects, including promoting cell survival by inducing the expression of the anti-apoptotic gene BCL2L1/BCL-X(L). To activate transcription, STAT6 requires interaction with coactivator proteins like p300 and CBP. A key physiological characteristic is its direct interaction with NF-κB, which underlies the synergistic transcriptional activation observed between the IL-4 and NF-κB signaling pathways. Clinically, its central role in IL-4-mediated responses makes it a molecule of interest in immune-related processes.
