Background
SREBF1 is a basic helix-loop-helix-leucine zipper transcription factor encoded by the SREBF1 gene and serves as a critical "master regulator" of lipid metabolism. In mammals, SREBF1 exists in two major isoforms—SREBP-1a and SREBP-1c—generated by distinct promoters, with SREBP-1c being predominant in metabolically active tissues such as the liver and adipose tissue, where it governs the synthesis of fatty acids and triglycerides. A unique feature of this protein is its mechanism for sensing cellular sterol levels within the endoplasmic reticulum membrane: when cholesterol is depleted, its N-terminal transcriptional active domain is released via proteolytic cleavage and translocates into the nucleus, where it binds to sterol regulatory elements in the promoters of target genes, activating the transcription of genes such as fatty acid synthase and acetyl-CoA carboxylase. Dysregulation of its function is closely associated with various metabolic diseases: hyperactivation promotes non-alcoholic fatty liver disease and insulin resistance, while variations in this gene have also been linked to traits such as obesity and dyslipidemia. Furthermore, through an interaction network with other transcription factors (such as LXR and ChREBP), this protein integrates signals from glucose and lipid metabolism to collectively maintain systemic energy homeostasis.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human kidney. Anti-SREBF1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
