Background
Sp7/Osterix is a ~45 kDa zinc-finger transcription factor of the Sp/KLF family that is indispensable for osteoblast lineage commitment and bone formation, acting downstream of Runx2 and cooperatively with BMP-6 to drive expression of osteogenic genes such as COL1A1, ALP, OCN, SATB2 and SOST, while its own transcription is induced by BMP-2, IGF-I and PTH and its activity is tuned by Akt- and p38 MAPK-mediated phosphorylation, ubiquitin–proteasome degradation and autoregulation via an OSX-bound tandem repeat in its promoter, and genetic or regulatory disruption of this short-lived nuclear protein underlies skeletal disorders including osteogenesis imperfecta and osteoporosis.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
