Background

The SP100 is a multifunctional protein primarily located within nuclear PML bodies (Promyelocytic Leukemia Protein bodies) and is a key component of these subnuclear structures. It exhibits a wide range of functions, acting as either a transcriptional co-activator or co-repressor in gene regulation, and through its interaction with HP1 family heterochromatin proteins, it links PML bodies to chromatin regions, thereby participating in chromatin dynamics and gene expression regulation. Physiologically, SP100 is involved in the regulation of cell growth, differentiation, and apoptosis, as well as in telomere maintenance, DNA damage response, and antiviral defense. Additionally, SP100 is an important autoantigen, and its antibodies serve as a critical serological marker for primary biliary cholangitis (PBC), an autoimmune disease. Furthermore, studies have revealed that SP100 plays complex and contradictory roles in various tumors, where changes in its expression levels are closely associated with the initiation, progression, and patient prognosis of different cancers, suggesting potential dual functions as either a tumor promoter or a tumor suppressor.

Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
