Background

SOCS2 is an important member of the SOCS family. As a classic negative feedback regulatory protein, it plays a critical inhibitory role in the JAK/STAT signaling pathway. The core function of SOCS2 is to specifically recognize and bind to phosphorylated tyrosine sites on target proteins through its SH2 domain, and it then serves as the substrate-recognition component of the E3 ubiquitin ligase complex (CRL5), mediating the ubiquitination and proteasomal degradation of target proteins. Studies have confirmed that SOCS2 is a major negative regulator of the growth hormone (GH) signaling pathway, and SOCS2 knockout mice exhibit a pronounced overgrowth (gigantism) phenotype, with body weight, body length, and bone length all increased by approximately 30–50%. In addition to the growth hormone receptor, SOCS2 can also target multiple substrates such as the erythropoietin receptor, leptin receptor, epidermal growth factor receptor, and insulin-like growth factor-1 receptor. SOCS2 is widely expressed in various human tissues and, beyond regulating growth, also participates in numerous physiological processes including central nervous system function, metabolic regulation, immune responses, and mammary gland development. Recent studies have found that SOCS2 is abnormally expressed in various tumors; for example, in hepatocellular carcinoma, SOCS2 expression is downregulated, and its overexpression can inhibit tumor cell proliferation, migration, and invasion, whereas in pancreatic cancer, upregulated SOCS2 can promote mitochondrial dysfunction and ferroptosis. Therefore, SOCS2 is considered an important molecule linking cytokine signaling regulation with tumor initiation and progression, holding potential value as a diagnostic and therapeutic target.
