Background
SNRPA1 (small nuclear ribonucleoprotein polypeptide A', also known as U2 snRNP A') is a core protein component of the spliceosomal U2 small nuclear ribonucleoprotein (snRNP) and plays a key role in pre-mRNA splicing. The human SNRPA1 gene is located at chromosome 15q26.3, and the encoded protein contains leucine-rich repeats (LRRs) and an RNA recognition motif, enabling it to bind the stem-loop structure of U2 snRNA and participate in the assembly of the U2-type prespliceosome and the catalytic step 2 spliceosome. Functionally, SNRPA1 not only supports constitutive splicing; recent studies have also revealed non-canonical regulatory roles in tumors: in breast cancer, SNRPA1 promotes a pro-metastatic exon inclusion program by binding specific RNA structural elements, enhancing lung colonization and invasive capacity; in non-small cell lung cancer, SNRPA1 is stabilized by METTL3/IGF2BP2-mediated m6A methylation and drives malignant phenotypes through interaction with TWIST1; in colorectal cancer, SNRPA1 acts as a partner protein of the long non-coding RNA RMRP, assisting MDM2-mediated p53 degradation and exerting oncogenic functions. In addition, SNRPA1 is indispensable for spermatogenesis, and loss of its function prevents spermatogonia from transitioning from proliferation to differentiation, being associated with human non-obstructive azoospermia. In the autoimmune field, SNRPA1 is a component of the U1/U2 snRNP autoantigen and is associated with autoantibody responses in systemic lupus erythematosus and mixed connective tissue disease.
