Background

SMC3 (Structural Maintenance of Chromosomes 3) is a crucial ATPase subunit of the cohesin complex, a ring-shaped protein assembly essential for sister chromatid cohesion, DNA repair, and the regulation of gene expression in eukaryotic cells. As a member of the Structural Maintenance of Chromosomes (SMC) family, SMC3 forms a heterodimer with SMC1 via their hinge domains, while their ATPase head domains interact with non-SMC subunits RAD21 (also known as SCC1) and either SA1/SA2 (STAG1/STAG2) to establish the topological embrace that encircles DNA strands. This structural configuration allows cohesin to hold replicated sister chromatids together from S-phase until anaphase, ensuring accurate chromosome segregation during cell division, thereby preventing aneuploidy and maintaining genomic stability. Beyond its canonical role in mitosis, SMC3-containing cohesin complexes are integral to chromatin architecture, facilitating long-range DNA looping and enhancer-promoter interactions that regulate transcription, and defects or mutations in the SMC3 gene are associated with developmental disorders such as Cornelia de Lange syndrome (CdLS) and various cancers, highlighting its pivotal role in both cellular physiology and human disease pathogenesis.
