Background
SMC1 (Structural Maintenance of Chromosomes 1) is a highly conserved ATPase subunit of the cohesin complex, which plays a pivotal role in maintaining chromosomal architecture and ensuring genomic stability in eukaryotic cells. As a core component of the ring-shaped cohesin structure, SMC1 dimerizes with SMC3 to form the structural backbone that topologically entraps sister chromatids, thereby facilitating sister chromatid cohesion from S-phase until anaphase onset to guarantee accurate chromosome segregation during cell division. Beyond its canonical function in mitosis and meiosis, SMC1 is critically involved in DNA double-strand break repair via homologous recombination, regulation of gene expression through chromatin looping, and the establishment of proper three-dimensional genome organization. Mutations or dysregulation of SMC1 are associated with severe developmental disorders known as cohesinopathies, such as Cornelia de Lange syndrome, and are also implicated in various cancers, highlighting its essential nature in both cellular physiology and human disease pathology.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon. Anti-SMC1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- SMC1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
