Background
SMARCAL1 (SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily A-like 1) is a DNA-stimulated ATPase and annealing helicase that travels with elongating replication forks and, upon detecting replication stress or DNA damage, catalyzes fork regression to form a protective “chicken-foot” structure and subsequent fork restoration, thereby stabilizing stalled forks, promoting template switching and homology-directed repair, and preventing MUS81-mediated double-strand breaks, while its tight regulation by RPA binding orientation and ATR-dependent phosphorylation of Ser-652 ensures controlled remodeling activity essential for maintaining genome stability, and germline mutations that abolish these functions cause the multisystem disorder Schimke immuno-osseous dysplasia.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon cancer. Anti-SMARCAL1 antibody was used at 1/50 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
