Background
SMARCA3, also known as HLTF (helicase-like transcription factor), is a member of the SWI/SNF chromatin remodeling family. However, unlike traditional remodeling complexes, it functions independently through its unique dual enzymatic activities—ATP-dependent helicase-like activity and E3 ubiquitin ligase activity. Its functions are multifaceted: in DNA replication and damage response, it catalyzes K63-linked polyubiquitination of PCNA protein, promoting replication fork reversal to repair damaged DNA; simultaneously, it also serves as a specific E3 ubiquitin ligase for histone H3K23, stabilizing chromatin structure by maintaining the repressive histone modification H3K9me3. SMARCA3 is widely expressed in various tissues, particularly in the nervous system where it binds to the p11/annexin A2 complex and participates in regulating behavioral responses to antidepressant drugs. Its dysregulated expression is closely associated with tumors: in colorectal cancer, approximately 40% of cases exhibit SMARCA3 silencing due to promoter hypermethylation of the HLTF gene, which is associated with chromatin opening, oncogene activation, and tumor progression, establishing its important role as a tumor suppressor.
