Background

SLC8A1, also known as NCX1, is a 70-kDa Na⁺/Ca²⁺ exchanger that resides primarily in the sarcolemma of cardiomyocytes, where it catalyzes the electrogenic counter-transport of 3 Na⁺ for 1 Ca²⁺ and thus functions as the dominant Ca²⁺ efflux pathway during diastole; its activity is allosterically regulated by intracellular Ca²⁺, inhibited by intracellular Na⁺ and the lipid PIP₂, and its dysfunction or overexpression contributes to arrhythmias, heart failure, and hypertension, while the gene contains multiple promoters giving rise to tissue-specific splice variants that differ only in their short regulatory N-terminal regions.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded rat cerebral cortex. Anti-SLC8A1 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
