Background

SLC40A1, also known as Ferroportin, is a crucial iron exporter protein that plays a vital role in iron homeostasis. It is primarily located in the plasma membrane of cells involved in iron metabolism, such as enterocytes, hepatocytes, and macrophages. In enterocytes, Ferroportin facilitates the transport of iron absorbed from the diet into the bloodstream. In hepatocytes and macrophages, it helps in the release of stored iron for systemic use. This protein contains multiple transmembrane domains and functions by moving iron ions from the intracellular space to the extracellular environment. Mutations in the SLC40A1 gene can lead to conditions like hemochromatosis, where iron overload occurs due to impaired iron export. Its regulation is also influenced by hepcidin, a hormone that binds to Ferroportin and induces its internalization and degradation, thereby controlling iron release from cells.

Immunocytochemistry

Anti- SLC40A1/Ferroportin antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
