Background

SLC12A2 (solute carrier family 12 member 2), also known as NKCC1 (Na⁺-K⁺-2Cl⁻ cotransporter-1), is a 1,215-residue, bumetanide-sensitive, electromeutral plasma-membrane protein that uses the inward Na⁺ gradient generated by Na⁺/K⁺-ATPase to drive uphill import of K⁺ and Cl⁻ into cells, thereby regulating ionic homeostasis, cell volume, epithelial salt movement, blood pressure, hearing, GABAergic inhibition, and neuronal excitability through dynamic phosphorylation by oxidative-stress-responsive kinases (e.g., SPAK/OSR1) and membrane trafficking in epithelia, neurons, vascular smooth muscle, immune cells, and secretory glands, with pathophysiological relevance to hypertension, deafness, neuropathic pain, and tumor cell proliferation.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
