Background

Skp2 (S-phase kinase-associated protein 2) is an F-box protein that serves as the substrate recognition component of the SCF (Skp1-Cul1-F-box) E3 ubiquitin ligase complex, playing a pivotal role in regulating cell cycle progression, particularly G1/S transition, through targeted proteolysis of key cell cycle regulators. As an oncoprotein frequently overexpressed in various human cancers, Skp2 specifically recognizes and promotes the ubiquitination and subsequent degradation of critical tumor suppressors including p27Kip1, p21Cip1, p57Kip2, and FOXO1, thereby driving cellular proliferation and inhibiting apoptosis. Beyond its canonical function in cell cycle control, Skp2 has emerged as a multifaceted regulator involved in DNA replication, metabolism, autophagy, and cancer metastasis through interactions with diverse substrates such as cyclin E, c-Myc, and Akt. The aberrant elevation of Skp2 expression, often resulting from genomic amplification, transcriptional upregulation, or impaired degradation, correlates with poor prognosis, tumor aggressiveness, and therapeutic resistance across multiple malignancies including breast, prostate, and lung cancers, making Skp2 an attractive therapeutic target for cancer intervention.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-Skp2 antibody was used at 1/100 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti-Skp2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
