Background
The SIGMAR1 protein, also known as the sigma-1 receptor, is a multifunctional chaperone protein composed of 223 amino acids with a molecular mass of 25.3 kDa. It is encoded by the SIGMAR1 gene and is ubiquitously expressed in various tissues, including the brain, heart, liver, and kidneys. SIGMAR1 is primarily located in the mitochondria-associated endoplasmic reticulum membrane (MAM), where it regulates calcium signaling, ion channel activity, and mitochondrial function. This protein plays a crucial role in cellular survival and has been implicated in several diseases, such as neurodegenerative disorders (e.g., amyotrophic lateral sclerosis and Alzheimer's disease), cardiovascular diseases, and certain cancers. Additionally, SIGMAR1 has been shown to have protective effects against neuronal and cardiac injuries.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- SIGMAR1 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
