Background
SHARPIN (Shank-associated RH domain-interacting protein) is a ~34 kDa ubiquitin-like adaptor that forms the LUBAC complex with HOIP and HOIL-1L to catalyze M1-linked linear ubiquitination of NEMO and other substrates, thereby activating canonical NF-κB signaling, suppressing TNF-α-induced apoptosis, modulating integrin activation, stabilizing β-catenin and ERα to drive Wnt/estrogen-dependent transcription, inhibiting ferroptosis via the p53/SLC7A11/GPX4 axis, promoting Rap1-mediated melanoma metastasis, and regulating Aβ phagocytosis and NLRP3 inflammasome activity in macrophages, with its dysregulation linked to chronic dermatitis, autoimmunity, and cancers of breast, stomach, and skin.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human kidney. Anti-SHARPIN antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in PC-3 cells. Anti-SHARPIN antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
